<p>PURPOSE:To accomplish mass proliferation of gentian as well as long-term preservation of the gene and chromosome types of gentian by putting the extirpated shoot apexes of gentian in a base medium containing plant growth hormone under rotary culture, specified temperature, illumination and rpm, and the resulting shoot primordia are put to static culture into seedlings. CONSTITUTION:The shoot apexes of gentian are transplanted to a base medium (e.g. conventional Murashige Skoog medium) containing plant growth hormone such as naphthaleneacetic acid or kinetin, and put to rotary culture at a temperature of 15-25 (pref. ca. 20) C, an illumination of 2000-10000 luxes and a rpm of 0.5-10 (pref. ca. 2). The resultant shoot primordia are cut e.g. into blocks of 3mm size, being transplanted to seedling medium prepared by incorporating the plant growth hormone with the base medium and put to static culture. Thereby, the gene and chromosome types of several kinds of gentian can be preserved for many yeats and gentian rich in nutrient can be proliferated in great quantities.</p> |
