GEL FOR FRACTIONATING AND PURIFYING RESTRICTION ENZYME

PURPOSE:To make it possible to specifically absorb a specific restriction enzyme in inexpensive production cost without immobilizing into a carrier by using a polysaccharide gel derived from seaweed as a gel for fractionating and purifying the restriction enzyme. CONSTITUTION:A polysaccharide gel, e.g. kappa-carrageenan gel or lambda-carrageenan gel, derived from a seaweed is used as a gel for fractionating and purifying restriction enzyme. In this case, kappa-carrageenan gel absorbs an enzyme of Hind III analog strongly and is weak in absorption of enzyme of EcolRI and PstI analog, while lambda-carrageenan absorbs an enzyme of BamHI analog specifically. The absorbed enzyme can be readily eluted by normally treating the enzyme with a buffer containing 0.5M sodium chloride.
