ALKALI PROTEASE PROMOTER, TERMINATOR FOR THE SAME PROTEASE, GENE-MANIFESTING UNIT COMPRISING THEREOF AND GENOME GENE OF ALKALI PROTEASE

摘要

NEW MATERIAL:An alkali protease promoter derived from genome gene of alkali protease of Aspergillus oryzae. EXAMPLE:The subject promoter having the base sequence expressed by formula I. USE:Used for a manifestation vector in host-vector system in mold, etc. PREPARATION:Chromosomal DNA is extracted from strain of Aspergillus oryzae and partially digested by a restriction enzyme, then introduced into a plasmid vector, thus the host is transformed to obtain a genomic library. Next, plasmid pAP017 containing about 6.5kb BglII fragment using oligonucleotide having the sequence expressed by formula II as a probe is obtained and the plasmid is subjected to NcoI digestion, treatment with T4DNA polymeasure, additon of EcoRI linker and FspI digestion, in turn, to afford the objective DNA fragment containing alkali protease promoter region.
