PURPOSE:To prepare a novel physiologically active polypeptide having an antitumor activity by culturing a microorganism transformed with a plasmid containing a DNA region coding the specific polypeptide. CONSTITUTION:A codon specifying amino acids constituting a human tumor necrosis factor(TNF) protein is selected and subjected to a chemical synthesis to prepare a human TNF gene (A). The component A is bound to a terminator, etc., and cloned in a vector to prepare a human TNF gene expression type plasmid (B) such as pTNF 401A. The component B is treated with a restriction enzyme to prepare an antitumor activity-having polypepetide-gene expression type plasmid pTNF 611 (C) containing a DNA of formula I coding the antitumor activity-having polypeptide. The component C is transduced into an Escherichia coli strain to prepare a transformant (D). The component D is aerobically cultured in a medium containing glucose, etc., at approximately 37 deg.C for 2-36hr, followed by purifying the cultured product by a centrifugal separation method, an electrophoresis method, etc., to produce the physiologically active polypepetide having an amino acid sequence of formula II and an amino terminal bonded to a Met. |
