NEW MATERIAL:The subject gene having a base sequence expressed by the formula. USE:For obtaining 2-halogenic acid dehalogenases. PREPARATION:A DNA is separated and purified from a microorganism having the ability to produce 2-halogenic acid dehalogenases and then cleaved with a restriction enzyme, etc. The resultant DNA fragment is subsequently joined to single strand of a suitable expression vector at adhesive terminal parts, etc., of the DNA chain and cyclized to afford a recombinant DNA vector, which is then ingressed into a replicative host microorganism. A microorganism holding a recombinant DNA vector is subsequently selected using 2-halogenic acid dehalogenases activity as an index to separate and purify the recombinant DNA vector from cultured microbial cells thereof. Thereby, the DNA which is the subject gene is collected. |
