NEW MATERIAL:Chondroitinases ACIII, C and BII. Action: the enzymes are lyases acting on the hexosaminide bonds of the saccharide chains of chondroitin sulfate and form a double bond between the 4 and 5 carbon atoms of uronic acid at both the ends of a broken section thereof; substrate specificity: the ACIII acts on hyaluronic acid to produce an unsaturated disaccharide and unsaturated oligosaccharides and also acts on the chondroitin sulfate to produce Di-OS, etc., but dose not Di-dis; the optimal pHs (50mM tris-acetic acid, tris-hydrochloric acid and glycine buffer solution, 37 deg.C): ACIII is a pH of 6-7; the optimal temperature for action (50mM tris-acetic acid, pH7): AC is 40 deg.C; inhibition and activation : the ACIII is activated with Ba<2+>. etc., and inhibited with Co<2+>, etc. USE:An enzyme used for more specifically breaking saccharide chains and useful for the research of the relation between the structure and activity of glucosamine glucan. PREPARATION:The above-mentioned enzyme-producing bacterium [preferably Flavobacterium Hp 102 strain (FERM 10206) belonging to the genus Flavobacterium produces the objective enzyme. |
