PURPOSE:To obtain a human protein by transforming blue-green algae cell using recombinant plasmid incorporated with the DNA sequence coding a polypeptide having the same amino acid sequence as that for human superoxide dismutase. CONSTITUTION:Firstly, a recombinant plasmid (or a DNA fragment capable of manifesting it) incorporated with the DNA sequence coding a polypeptide having virtually the same amino acid sequence as that for human superoxide dismutase (h-SOD) is synthesized. Second, using this recombinant plasmid (or the DNA fragment capable of manifesting it), blue-green algae put to culture in a medium to manifest polypeptide. Thence, cells are collected from the resulting cultured product by e.g. centrifugation and then ground, and the resultant polypeptide is separated by means of e.g. salting-out and recovered. The polypeptide thus obtained is useful for e.g. the pharmaceuticals for the therapy or prevention of chronic articular rheumatism, osteoarthritis, etc. |
