PURPOSE: To maximize viability of transplantable tissue after cryopreservation by putting a cell into a nutrient medium and incubating the cell at a specific temperature and time. CONSTITUTION: A transplantable tissue is incubated in a nutrient medium at 27-42 deg.C for 5-24hr (preferably at 37 deg.C for 6hr) in a incubator or shaking water bath prior to cryopreservation. Thereby, a cell or the tissue is revitalized and viability and functional capacity of cell in thawing are improved. Further, cryopreserved tissue or cell is survived in thawing and transplanting and functionalizing ability can be improved by the above method. The nutrient medium used includes an equilibrium tissue culture medium, a simple phosphate buffer salt solution (in which a nutrient such as glucose is supplemented),etc. and a protein suspension (e.g. serum or artificial serum) may exist in the medium. |
