PURPOSE: To obtain a pharmaceutical composition, in which an active substance is uniformly dispersed, enabling long-term controlled release by dry-mixing microparticles of a copolymer of lactic acid with glycolic acid with a salt of a peptide, precompressing and preheating the resultant powder mixture, extruding the mixture and pulverizing the extruded mixture at a low temperature. CONSTITUTION: This pharmaceutical composition is obtained by dry-mixing microparticles of a copolymer of lactic acid and glycolic acid having <=200μm, preferably <=180μm average particle size (preferably composed of 45-90mol% of the former and 55-10mol% of the latter) with pamoate, tannate, stearate or palmitate of a natural or synthetic peptide, continuously compressing the resultant powder mixture and continuously heating the mixture to about 80 deg.C and extruding the mixture at 80-100 deg.C under 50-500kg/m<2> pressure and pulverizing the extruded mixture at a low temperature and selecting and collecting microparticles having <=200μm particle size. The peptide includes luteinizing hormone-releasing hormone, somatostatin, calcitonin, growth hormone-release hormone.
