NOVEL TISSUE PLASMINOGEN ACTIVATING FACTOR

PURPOSE:To produce t-PA having a finger domain and/or a cringle 1 domain negatively charged by transforming a host cell with a manifesting vector containing DNA coding said t-PA. CONSTITUTION:Host cell is transformed using a manifestation vector containing DNA coding human tissue plasminogen activating factor (t-PA) having a finger domain and/or a cringle 1 domain having negative electric charge. Said transformed host cell is cultured in a medium and generated t-PA is collected from the cultured substance. Resultant t-PA is useful as fibrinolytic agent for the treatment of vascular disease. Besides, cultivation of said transformant is preferably performed at pH about 7-7.5 for about 5-50hr.
