PURPOSE:To determine a substrate up to high conc. by calculating the unknown concn. of the substrate using the output values to a plurality of standard substrates. CONSTITUTION:A buffer solution 1 is fed in a flow rate of 1.0 ml/min using a pump 2 for high performance chromatography. Subsequently, the buffer solution 1 containing a specimen completed in measurement is collected in a waste solution bottle 11. A measured value is digitized to be taken in a computer 10 and a base current value is subtracted from the peak current value obtained when the specimen passes through an electrode to calculate a detected current value. At this time, the output values to a plurality of standard substrates are used to calculate approximate values to Km and V (Km and V are constant) in formula I (wherein I is an output value and S is the concn. of a substrate) and, further, the optimum values of Km and V are calculated from the formula and the unknown concn. of a substrate is calculated from the formula I for which the optimum values are substituted. |
