NEW MATERIAL:A DNA capable of coding a protein which inactivates the function of tabtoxin. USE:For creating plants resistant to Pseudomonas syringae pv. tabaci. PREPARATION:For example, Pseudomonas syringae pv. tabaci is cultured and collected and a chromosomic DNA is then separated by a conventional method and treated with a restriction enzyme to prepare a chromosomic DNA fragment. On the other hand, a plasmid pUC 13 of Escherichia coil is treated with a restriction enzyme and subsequently linked to the chromosomic DNA fragment of the Pseudomonas syringae pv. tabaci and then inserted into Escherichia coli to carry out transformation. The resultant transformant is subsequently cultured in an agar culture medium containing ampicillin to culture the grown colony in a culture medium containing tabtoxin and thiamine. Thereby, a colony having tolerance to the ampicillin and tabtoxin is collected to separate a plasmid according to a conventional method. The resultant plasmid is further treated with a restriction enzyme to afford a DNA capable of coding a protein capable of inactivating the function of the tabtoxin. |
