The invention disclosed herein provides ribosomal frameshift signal sequences which cause a ribosome arriving at the signal sequences during translation, to slip back by at least one nucleotide to result in the decoding of the subsequent downstream RNA sequence into a different reading frame. The invention also provides a nucleotide construct which contains a gene for a desired primary polypeptide together with a gene for a secondary polypeptide with said ribosomal frameshift signal sequence located between said genes, whereby translation of the secondary polypeptide gene depends on whether or not frameshifting has occurred. Also provided are recombinant cloning vectors, replicable expression vectors, recombinant microorganisms and cell cultures containing the nucleotide constructs. Also provided are protein expression products expressed by use of the nucleotide constructs and methods utilising the nucleotide constructs. |
