<p>Cpds. and their prepn. (I) R1=H or Me R2=isopropyl or isobutyl a=double or single bond (II) Acid addn. salts of (I). Adrenolytics with pressor and depressor activity. I (R1=Me) also have anti-serotonin activity. C5H5N (20 ml) was added dropwise to 9,10-dihydrolysergic acid chloride (6 g) in MeCN (30 ml) and DMF (30 ml), followed by (IV) (R2=Bui, 3.44 g), stirred 1.5 hrs. at 0 deg., decompd. with 2N-HCl (40 ml) and poured into 2N-NaOH (200 ml). The ppt. was extd. with 10% EtOH in CH2Cl2 (5 X 200 ml), the extract washed with d. NH4OH (100 ml) and brine, decolourised, evapd., and 2 times evapd. with PhMe (40 ml). The residue was chromat. on II-III Al2O3, eluting with 6% MeOH in CH2Cl2, recryst. from EtOA, and pptd. from AcOH with Et2O, giving 9,10-dihydroergonine, m.p 218-220 deg.</p> |
