PURPOSE:To enable efficient obtaining of an indole alkaloid in a large amount by carrying out tissue culture of tissues from a plant capable of producing the indole alkaloid in a culture medium containing fructose as a saccharide source. CONSTITUTION:A cultivar Cathranthus roseus var. Little.Bright.Eye, etc., of a plant of the genus Lochnera is bedded on a solid Murashige.Skoog culture medium containing agar and cultured at 10 to 35 deg.C for 7 to 30 days to provide calli. On the other hand, 2 to 5wt.% fructose is blended with 0.1muM to 100mM inorganic ingredient, such a s Ca, 0.01 to 10muM phytohormone, such as indoleacetic acids, 0.1 to 100mg/l each of vitamins, such as biotin or glycine, and amino acids and added to the Murashige.Skoog culture medium to afford a liquid culture medium for tissue culture. The aforementioned calli are then inoculated into the abovementioned culture medium and cultured at 10 to 35 deg.C. After completing the culturing. calli are separated by a decantation method or filtration, etc., to afford calli, which are subsequently extracted and purified with methanol, etc., to produce an indole alkaloid, such as catharanthine. |
