The stability of cloning vectors for use in gram-positive hosts, both structural and in many cass segregational stability, can be improved by inserting into the vector a nucleic acid sequence which acts as a lagging strand origin of replication (LSO) in the host organism. Accordingly, the invention encompasses a method of stabilizing cloning vectors by insertion of an LSO into the nucleic acid sequence of the vector and the cloning vectors thus stabilized. In particular, vectors carrying the lagging strand origin from the plasmid pUB110 exhibit improved stability when resident in B. subtilis and other gram-positive hosts. |
