PURPOSE: To provide substantially the same biological activity as hirudin by combining a functional group of a supporting material directly with hirudin or proteins of hirudin derivatives utilizing anhydrous or aqueous combining reactions. CONSTITUTION: To the active functional groups of a supporting material such as hydroxyl group, carboxyl group or amino group, hirudin or proteins of hirudin derivatives is combined by functional groups of amino acid residues. Here, materials and protein include natural products, genetic derivatives and synthetic substances. Methods of combining depend upon factors such as useful functional groups of the support materials, the combining positions of proteins, biological activity of materials obtained, and selectivity and efficiency of combining reactions. There are two methods; one performed under an anhydrous or aqueous condition and the other using dual active protein cross-linking reagents. During this combining process, the active functional group of the amino acid residue is protected. |
