<p>The present invention is in an immunoassay system and process for detecting analytes and is illustrated for detecting specific antibody presence in a specimen in a short period of time as in a non-laboratory setting, such as a doctor's office. The system includes a support whereon one or more dots of a specific purified antigen to be tested for are spotted and the area around that spot or spots is preferably blocked. The support is for sequential incubations of approximately four (4) minutes each in reagent vessels containing: a specimen consisting of a diluent and a drop or drops of plasma, serum or whole blood; a high ionic strength wash solution; an enzyme conjugate; a second wash solution containing, depending upon the test procedure, a high or low ionic strength solution; and an enzyme substrate that produces a coloration of the antigen-antibody binding site of an antibody-antigen. The system of the present invention employs a high ionic strength solution as the first wash solution of from 0.25 M to saturation for removing interfering substances, presumably those substances having lower binding affinities, and for increasing sensitivity. The system reagents are preferably tabletized in the manufacture and are reconstituted just prior to use. Applicant has successfully combined dry and tabletized, as the enzyme substrate reactant, a compound of 5-bromo-4-chloro-3-indolyl phosphate (BCIP) and P-nitro blue tetrazolium chloride (NBT).</p> |
