PURPOSE:To produce a mixture of the subject sugars either by aerobically culturing a microbial strain belonging to genus Klebsiella in a medium containing sucrose as main carbon source or by reacting the obtained microbial cells. CONSTITUTION:Klebsiella oxytoca S-61 (FFRN P-10663) belonging to genus Klebsiella is cultured in advance and inoculated to a medium produced by adding sucrose as a main carbon source, a nitrogen compound such as peptone and further yeast extract, phosphoric acid salt, etc., to a medium. The microorganism is cultured under aerobic condition at 20-37 deg.C under shaking to accumulate the objective palatinose and trehalose in the medium. As an alternative method, the microbial cell produced by the cultivation or treated cell is made to react with sucrose. The culture liquid or a reaction liquid of S-61 cell is subjected e.g. to centrifugal separation to remove the microbial cells. The obtained supernatant liquid or filtrate is desalted and concentrated to obtain a mixed syrup of both sugars. Palatinose and trehalose are separated in purified state from the syrup by conventional technique. |
