PURPOSE:To simultaneously determine the number and position of a double bond, by esterifying unsaturated fatty acid by reacting the same with a trialkylsililation agent without performing chemical treatment while subjecting the esterified product to a gas chromatography mass spectrometry (GCMS method). CONSTITUTION:The carboxyl group of unsaturated fatty acid in a specimen is reacted with a trialkylsililation agent (a) to form a trialkylsilil ester derivative of unsaturated fatty acid which is, in turn, subjected to a GCMS method based on a chemical ionization method using isobutane, methane or ammonia as an ion source and the number and position of the double bond in unsaturated fatty acid is determined from the mass spectrum of a psuedo-molecule ion and a fragment ion. As the trialkylsililation agent, a mixture of trimethylsilil chrolide and hexamethyl disilazane is pref. used and reaction is performed in an inert anhydrous org. solvent in the presence of an acid acceptor (base) such as pyridine or acetonitrile.
