摘要 PURPOSE:To enable the rapid and accurate quantitative analysis of bilirubin in serum, by measuring the reduction in the yellow color of bilirubin or the amount of residual K3[Fe(CN)6] according to a color developing method by oxidizing bilirubin by reacting K3[Fe(CN)6] with a specimen. CONSTITUTION:0.03-1.0mM/test of K3[Fe(CN)6] is added to a solution prepared by adding about 3ml of a 0.1M phosphate buffer solution (pH 7.0) containing 0.25% of sodium cholate to 100mul of serum specimen to perform reaction at 25- 45 deg.C for 5-20min and optical density Es is measured at a wavelength of 440- 480nm, pref., 460nm. Separately, the optical density Esb of a serum blank having no reagent added thereto is measured and the bilirubin concn. of the serum specimen is accurately measured by the formula obtained from reference optical density Est and the optical density Est(B) of a reference blank by using serum of which the bilirubin concn. is known as reference. The amount of a residual reagent may be measured by adding a color forming reagent.
