PURPOSE:To obtain in high yield the title enzyme of high agar degradation ability by making a culture of specific bacteria classified as Vibrio sp. in a short time with easy control. CONSTITUTION:The objective enzyme produced by microorganisms classified as Vibrio sp., e.g. Vibrio sp. SCRC-8007 (FERM P-10560) having the following characteristics: A) degradable for agar-agar and pectin, but virtually inert to mannan, dextran, alginic acid and cellulose, B) optimum pH for action; ca. 6-8, and C) optimum temperature for action: ca. 30-50 deg.C. To produce the present enzyme, the title bacteria is put to culture in a medium containing (1) agar-agar as carbon source, (2) if needed, nitrogen source such as yeast extract, and (3) natural seawater, etc., pref. at 15-25 deg.C and pH of 7-8 for 12-48hr, and from the resultant culture solution the objective enzyme is separated and purified. A degradation product of agar-agar can be obtained from the above culture solution through the separation and purification process for sugar. |
