NEW MATERIAL:A cytotoxic active substance, purified from human LAK cell and having the following physico-chemical properties. Molecular weight; about 35,000 (gel filtration method) and about 41,000 (SDS-electrophoresis). Having common antigenicity with lymphotoxin without requiring calcium ions in the target cytotoxicity. Causing fragmentation of DNA in expression of cytotoxic activity. USE:A carcinostatic agent. PREPARATION:For example, a monocyte collected from normal human peripheral blood is treated by a conventional method and induced into an LAK cell having the LAK activity, which is then destroyed by N2 cavitation and centrifuged to remove the nucleus. Ultrasonic crushing is carried out in a buffer solution to dialyze the cell. Ion exchange column chromatography and gel filtration are successively carried out to purify the dialyzate and afford the objective substance having cytotoxicity. |
