NEW MATERIAL:A hemolytic protein originated from the fruit body of Pleurotus ostreatus, having a molecular weight of 18kDa and an isoelectric point of 6.3, bonding to a biomembrane containing sphingomyelin and exhibiting strong hemolytic action by synergistically acting with a hemolytic protein originated from the fruit body of Pleurotus ostreatus and having a molecular weight of 48kDa and an isoelectric point of 6.3. USE:A remedy for cancer, etc., and a reagent for detecting sphingomyelin. PREPARATION:The objective hemolytic protein of Pleurotus ostreatus can be produced e.g. by disintegrating the fruit body of Pleurotus ostreatus together with a tris-HCl buffer solution (pH8.0) with a blender, etc., centrifuging the homogenate to collect the supernatant liquid, adding ammonium sulfate to the liquid, removing the produced precipitate, desalting the supernatant, passing through an ion exchange column, collecting unabsorbed fraction and successively purifying by hydroxyapatite column chromatography and gel-filtration. |
