PURPOSE:To obtain NAD synthase useful as an enzyme for diagnosis, stabler than conventionally well-known NAD synthase by culturing a bacterium belonging to the genus Bacillus, capable of producing NAD synthase and collecting the NAD synthase from the culture solution. CONSTITUTION:For example, Bacillus licheniformis B-0844 (FERM P-6809) is prepared as a bacterium capable of producing NAD synthase. The bacterium is cultured in a medium containing a carbon source such as glucose, a nitrogen source such as corn or soybean flour, and, if necessary, an inorganic salt. The culture is carried out at 26-50 deg.C for about 15-40 hours. Then the bacterium is collected by a means such as filtering or centrifuging the culture mixture and the cell is destroyed by a mechanical means such as ultrasonic treatment. Then the NAD synthase is precipitated by a method such as subjecting the prepared solution containing the NAD synthase to salting-out with ammonium sulfate and the NAD synthase is collected. |
