摘要 |
PURPOSE:To readily and effectively mass-produce a synthetic gene coding an amino acid sequence of hormone bond range of human growth hormone receptor, having two or more restriction enzyme recognition sites wherein a site recognized by one restriction enzyme exists at only one partion in the synthetic gene, by coding a specific amino acid sequence. CONSTITUTION:A synthetic oligonucleotide is subjected to bond reaction in the presence of ATP to give a synthetic gene (A) to code an amino acid sequence of hormone bond range of human growth hormone receptor which codes a natural amino acid sequence but has a nucleotide sequence different from that of natural gene: has two or more restriction enzyme recognition sites wherein a site recognized by one restriction enzyme exists at only one partion in the synthetic gene: not contains direct repeat of >=7 bases in arbitrary 60 nucleotide chains in nucleotide sequence: has restriction enzyme of 6 base pair palindrome recognition type enzyme selected from MluI, AccIII, SacI, etc. Then a plasmid obtained by transducing the component A is transformed to give Saccharomyces cerevisiae strain (B) is cultured in a medium to produce a protein containing a hormone bond range of human growth hormone receptor. |
