PURPOSE:To obtain a new polygalactosamine decomposition enzyme (polygalactosaminidase) A-4 from a cultured product by culturing a microbial strain belonging to genus Bacillus and capable of producing a polygalactosamine decomposition enzyme A-4. CONSTITUTION:A microbial strain capable of producing the above decomposition enzyme A-4 [e.g. Bacillus sp. A-4 (FERM P-10473) separated from soil] is cultured by the following procedures. The strain is cultured in a medium containing proper amounts of C source, N source, inorganic substances and other nutrient sources, preferably e.g. containing 0.5% of polygalactosamine, 0.5% of yeast extract, 0.05% of KCl, 0.1% of K2HPO4, 0.05% of MgSO4.7H2O and 0.01% of FeSO4 at 20-40 deg.C and starting pH of about 6-8 by shaking culture or submerged agitation culture. The objective new polygalactosamine decomposition enzyme A-4 capable of hydrolyzing polygalactosamine to produce oligogalactosamine can be separated from the medium. |
