<p>PURPOSE:To obtain novel alpha-amidated enzyme having C-end derived from human thyroid gland and biologically active fragment of said enzyme able to translate a protein or peptide expressed by specific formula to an alpha-amidated protein or peptide having C-end expressed by other specific formula. CONSTITUTION:A protein or peptide expressed by formula I is amidated to present new alpha-amidated enzyme having above-mentioned C-end able to be translated to formula II. To obtain said enzyme, at first, two species of cDNA of new alpha-amidated enzyme having C-end are isolated from human thyroid gland. Next, protein exhibiting activity to alpha-amidated enzyme having C-end is manifested in a large amount in escherichia coli and zooblast (COSI) by using said cDNA. Therefore, enzyme having activity to alpha-amidated enzyme having C-end is able to be produced in a large amount by using novel cDNA of alpha- amidated enzyme having C-end. Furthermore, industrially useful amidated peptide is able to be produced in a large amount and in a low cost by using said enzyme.</p> |
