<p>PURPOSE:To prepare a large amount of uniform indefinite embryo with simple operation in high efficiency by inducing a callus from a tissue piece of horseradish in an MS medium containing phytohormones, isolating the induced callus and classifying the product. CONSTITUTION:A tissue piece 3 having a proper size is cut out from a leaf 1 or stalk 2 of horseradish, sterilized and placed on a solid MS medium 4 containing a gelatinization agent such as agar added with phytohormones. The tissue piece is cultured in the medium 4 to induce a callus 8 (a cluster of cells) by dedifferentiation. The callus 8 is transferred to a liquid MS medium 9 containing the above phytohormones and cultured under shaking to obtain isolated and suspended cells 10, The suspended cells 10 are filtered with a filter 11, etc., to uniformize the particle size, added to a liquid MS medium 12 free from said phytohormone and cultured under shaking to obtain the objective indefinite embryo 5 with simple operation in high efficiency.</p> |
