PURPOSE:To discriminate change of base sequence in high accuracy by cleaving DNA multiplied by PCR method with a specific restriction enzyme specific to HLA antigen allospecificity, fractionating length of the fragment by electrophoresis and detecting. CONSTITUTION:DNA amplified by PCR method is cleft with a restriction enzyme specific to HLA (leukocyte membrane antigen) antigen allospecificity. Then length of the cleft fraction is fractionated by electrophoresis, detected and type of HLA is discriminated by difference in the length. Restriction enzyme ApaI, BssHII, DdeI, FokI, HacIII, HhaI, HinfI, KpnI, MaeI, MboII, SacII, SauzAI, ScrFI, etc., may be cited as the concrete example of the restriction enzyme. A HLA type discriminating diagnosticum comprising a restriction enzyme specific to HLA antigen allospecificity is prepared. |
