NEW MATERIAL:Monoclonal antibodies TM-A54, TM-A59, TM-A65, TM-A73, TM-A31, which specifically bind to a human originating thrombin-binding substance specifically to enhance the activation of protein C by bonding to thrombin. USE:Purification of the above-stated thrombin-binding substances (abbreviated to TM) in high purity and immunological assays. PREPARATION:Antibody-producing cells from an animal immunized with TM are fused with myeloma cells, the hybridoma is selectively proliferated to retrieve the antibody-producing hybridomas. The monoclonal antibody in the culture mixture of the antibody-producing hybridoma recognizes the site binding to TM thrombin and the desired hybridoma is cloned by investigating whether it recognizes the structural change of TM caused by calcium ions or the change in molecular weight by enzymatic treatment.
