PRODUCTION OF NON-CONSTITUTIVE PROTEIN OF JAPANESE ENCEPHALITIS VIRUS

PURPOSE:To obtain the subject substance useful as a Japanese encephalitis vaccine, etc., on an industrial scale at a low cost by infecting an insect cell with a recombinant containing a cDNA coding a non-constitutive protein of Japanese encephalitis virus in a genom region non-essential to the proliferation of a baculovirus and culturing the virus. CONSTITUTION:Japanese encephalitis virus is extracted with phenol to collect virus genom RNA, the RNA is treated with a transcriptase and the prepared cDNA is treated with a restriction enzyme, linked to a vector and inserted into E.coli to effect the transformation of the cell. The transformed cell is screened to select a clone containing cDNA coding a non-constitutive protein of Japanese encephalitis virus. A part or total of the cDNA is integrated into a genom region non-essential to the proliferation of baculovirus and the recombinant baculovirus is infected to an insect cell and cultured to obtain the expressed non-constitutive protein of Japanese encephalitis virus.
