PURPOSE:To obtain as bile acid sulfate sulfatase capable of specifically hydrolyzing a sulfuric ester part of a bile acid 3alpha-sulfate by culturing a bacterium belonging to Pseudomonas.testosteroni in a culture medium containing bile acid. CONSTITUTION:An enzyme, capable of acting on bile acid 3alpha-sulfate, hydrolyzing the sulfuric ester part, reversing the binding position of the OH group from the alpha- to the beta-position and forming 3beta-hydroxybile acid and having substrate specificity of acting on the free type bile acid 3alpha-sulfate and glycine conjugated type and taurine conjugated type bile acid 3alpha-sulfates, the optimum pH region of pH8.5+ or -0.5 and optimum temperature of 35+ or -5 deg.C and about 100000 molecular weight is used as a bile acid sulfate sulfatase. The above-mentioned enzyme is obtained by culturing Pseudomonas.testosteroni (preferably ATCC11996) in a culture medium containing bile acid under aerobic conditions at 24-34 deg.C for 12-24hr and extracting the objective enzyme from the microbial cells. |
