PURPOSE:To enable accurate determination in a short time by adding isocitrate dehydrogenase to a specimen and carrying out enzymic reaction. CONSTITUTION:Isocitrate dehydrogenase is dissolved in a buffer solution containing nicotinamide adenine dinucleotide(NAD)P<+>, isocitric acid, etc., to provide (A) an enzymic solution, which is then added to (B) a specimen, such as humor, containing glycol ether diaminetetraacetic acid (GEDTA) which is an Mn ion chelating agent added thereto and subjected to enzymic reaction for a desired time to afford (C) a reaction solution. An Mg ion chelating agent or adenosine triphosphate(ATP) or both as a reaction terminator (E) are added to the solution (C) to terminate the reaction. Mg is then determined by measuring NAD(P)H formed in the terminated reaction solution with an absorbance at 340nm wavelength.
