NEW MATERIAL:Immune suppression factors I and II having each 40,000+ or -5,000 and 10,000+ or -5,000 molecular weight and LAK(Lymphokine Activated Killer) induction suppressing activity. USE:An immunosuppresive agent. PREPARATION:For example, TUH-1 cell established from a human acute leukemia cell is subcultured and then centrifuged to collect the cell and the cell is inoculated into a culture medium to which a stimulating substance (e.g., Escherichia coli-derived lipopolysaccharide) activating differentiation of monocyte or function of macrophage is added and the cell is subjected to spinner culture in a temperature-controlled culture vessel containing 5% CO2 at 37 deg.C for 19hr. Then the culture medium is centrifuged to remove the cell and subjected to filtration sterilization with a membrane filter and the filtrate is concentrated by an ultrafiltration membrane and passed though a high speed liquid gel filtration column and fractionated to provide the immune suppression factors I and II. |
