NEW MATERIAL:A hematopoiesis controlling factor produced by a macrophage and having the following characteristics: Molecular weight, 40,000+ or -4,000 (by SDS-PACE and gel filtration method); chemical properties, absorbing into a silica gel, etc., in 10mM phosphoric acid buffer (pH7.0) and losing biological properties at a ratio of >=90% by 0.25% trypsinization and heat treatment at 56 deg.C for 30min; biological properties, supporting proliferation of IL3-dependent cell and suppressing proliferation of GM-CSF and G-CSF-dependent cell. USE:A remedy for blood cell disease and leukemia. PREPARATION:For example, a thioglycolate culture medium is inoculated into an abdominal cavity of mouse and peritoneal exudate liquid is collected and mononuclear cell in the exudate liquid is separated and suspended in a culture medium and cultured in carbon dioxide incubator and the culture medium is centrifuged to collect a supernatant, which is then fractionated by a gel filtration column chromatography to provide the hematopoiesis controlling factor. |
