Immobilized enzyme and a method for application thereof.

Porous chitosan beads are used as carriers for immobilization of polynucleotide phosphorylase (PNPase). Immobilization of PNPase to the porous chitosan bead carrier can be effected by contacting the enzyme with the carrier in water or in a buffer solution such as phosphate or borate buffer solution, preferably at room temperature. The thus obtained immobilized enzyme is contacted with a solution of nucleoside, e.g. cytidine-5 min -diphosphate or inosine-5 min -diphosphate, to produce the corresponding polynucleotide, e.g. poly C or poly I. The yield of polynucleotide is thus improved and the costs reduced. Also, the reaction can be conducted by a continuous process and the chain length of the polynucleotide can be controlled.
