PURPOSE:To obtain the title human lysozyme with high specific activity by replacing the amino acid at specific site in the amino acid sequence of natural- type human lysozyme by other amino acid. CONSTITUTION:Variant human lysozyme gene coding the polypeptide with valine at the 110 site on the amino acid sequence of natural-type human lysozyme replaced by asparagine is prepared. Thence, a manifestation vector (e.g., plasmid pERI 8866) is prepared, which contains the above variant human lysozyme gene and nucleotide sequence coding signal peptide and is capable of autonomous replication in eukaryote. Using this manifestation vector, variant human lysozyme is produced, and secretable host cells are transformed, and the resulting transformant is put to culture. The culture supernatant separated from the resultant cultured product and the variant human lysozyme in the fungi are then purified and isolated. Said host cell is pref.
