NEW MATERIAL:alpha-Amylase TF-35 having the following properties. Action, hydrolyzing a straight-chain structure composed of alpha-1,4-glucopyranoside bond of amylopectin, amylose, starch, etc., to form maltose; substrate specificity, able to hydrolyze a straight-chain malto-oligosaccharide, etc., composed of alpha-1,4-glucopyranoside bond and having a polymerization degree of higher than maltotriose but unable to hydrolyze pullulan, etc.; optimum pH, 5.5-6; optimum working temperature, 60 deg.C; inhibition, activation and stabilization, inhibited by Cu, etc., and stabilized by Ca; molecular weight (gel-filtration), 13,000+ or -2,000; isoelectric point (chromatofocusing process), 5.2; etc. USE:Useful for the production of maltose from starch in high productivity at a low cost. PREPARATION:The objective enzyme can be separated from a cultured product of a microbial strain belonging to genus Thermomonospora and capable of producing alpha-amylase TF-35 [preferably Thermomonospora viridis (FERM P-10339)]. |
