Disclosed are methods for determining antibodies against the pre-S regions contained in the L-protein and the M-protein in hepatitis B virus (HBV) env proteins by enzyme immunoassays, in which nonspecific reactions can be depressed by pre-treatment of serum with an antigenic protein produced by a unicellular organism and by contacting an antigenic protein produced by a unicellular organism, whereby all anti-pre-S1 and anti-pre-S2 antibodies in human sera and the sera of experimental animals can be highly sensitively determined without the use of radioactive materials.
