NEW MATERIAL:An osteoblast proliferation factor which is purified from human or mammalian bones and contains an active ingredient having the following properties. Isoelectric point; pH6.1-7.5. Eluting position in filtering with a natural gel filter medium) position of 25000-15000dal relative average molecular weight; eluting position in gel filtration under acidic conditions; position of 10000-4000dal relative average molecular weight. Stability; stable to heat treatment at 100 deg.C for 5min and reduction treatment with dithiothreitol. USE:Useful for treating osteoporosis, intractable fracture, etc., reagent for diagnosing and measuring diseases thereof, research for bone metabolism, etc. PREPARATION:A bone substrate of human femoral head is initially crushed to provide a bone meal, which is subjected to operation of freeze-drying, washing, defatting, dialytic and removing treatment of giant protein molecules, etc., to afford a Crude low-molecular weight protein. The resultant protein is then successively subjected to filtration with a natural gel filter medium and gel filtration under acidic conditions to collect an active fraction, which is subsequently purified. |
