PURPOSE:To obtain flagella, such as salmonella or Escherichia coli, in high purity and yield in simple operation by culturing a bacterium in a liquid culture medium containing an additive capable of forming a conjugate with proteins and collecting the resultant flagella. CONSTITUTION:A bacterium, such as salmonella or Escherichia coli, having flagella is cultured in a liquid culture medium containing an additive capable of forming a conjugate with proteins thereof by shaking culture for, e.g., 3-24hr. Polyethylene glycol or a derivative thereof is preferred as the above-mentioned additive and the molecular weight is preferably about 3000-50000. The afore- mentioned culture is then centrifuged to provide precipitates containing microbial cells and flagella. Water is subsequently added to dilute the above-mentioned precipitates and the resultant mixture is separated into a supernatant liquid containing suspended flagella and precipitates (the microbial cells). The objective high-purity flagella are then collected from the supernatant liquid in high yield. |
