NEW MATERIAL:Lymphokine-activating killer cell induction suppressing factor LAKSF having a molecular weight of 68000+ or -5000 and an isoelectric point of pH6.8+ or -0.5. USE:An immunosuppressing agent. PREPARATION:LAKSF can be produced e.g., by collecting peripheral blood of normal person, separating monocyte in high purity by continuous centrifugal separation, culturing the cell at 37 deg.C for 16hr in a medium containing E.coli- originated lipopolysaccharide as a stimulator, removing the monocyte by centrifugal separation, filtering and sterilizing the liquid with a membrane filter having a pore diameter of 0.45mum, concentrating the obtained crude LAKSF with an ultrafiltration membrane, passing through a column packed with crosslinked dextran gel to effect the fractionation and purifying the objective fraction by ultrafiltration and high-performance liquid chromatography. |
