PURPOSE:To increase producing amount of protein coded by heterogene by transforming host yeast having deteriorated protein phosphate enzyme activity dependent on cyclic AMP with manifestation vector having promoter derived from glycotic system and said heterogene, then culturing said transformant. CONSTITUTION:A mutant of enzyme gene relating to the above-mentioned host yeast, e.g., so-called 'cAMP path', e.g., 2-defect strain such as ras2, etc., is prepared. Then, said host yeast is transformed by an enzyme constructing a path generating pyruvic acid from a promoter derived from glycotic system, i.e., glucose, e.g., a promoter of gene coding pyruvate kinase, etc., and a manifestation vector having heterogene. Besides, e.g., a gene coding apoprotein AI, etc., is shown as said heterogene. Next, resultant the above-mentioned transformant is cultured by conventional method to afford the aimed protein coded by the above-mentioned heterogene. |
