PURPOSE:To provide a DNA fragment having a specific base sequence, exhibiting good promoter activity in yeast cell and advantageously usable as an expression vector capable of expressing various kinds of heterogenes. CONSTITUTION:The objective DNA fragment is expressed by the base sequence of formula at most and exhibits good promoter activity in yeast cell. The DNA fragment can be prepared e.g., by the following process. A chromosome DNA prepared from S.cerevisiae DKD-5D-H strain by conventional process is partially decomposed with a restriction enzyme to obtain individual DNA fragments. The DNA fragments are introduced into the nicked site of a vector and the DKD-5D-H strain is transformed with the obtained plasmid. A transformant having improved resistance against heavy metals is screened from the obtained transformants and the objective DNA fragment is prepared from the transformant. |
