<p>PURPOSE:To surely form a large amount of tuber of potato by adding ascorbic acid and a cytokinin compound as potato tuber formation inducers to a medium of sterile shoot. CONSTITUTION:A stem fragment containing a terminal bud or a node bred by shoot apex point culture or rooting and transferring method of potato plant is reared in a tissue culture medium (e.g., Linsmaier & Skoog medium) for about four weeks to give a sterile shoot. Then the medium of the sterile shoot is incorporated with 100-5,000ppm ascorbic acid and 0.5-10ppm cytokinin compound (e.g., kinetin) potato tuber formation inducers and the sterile shoot is cultured for 2-4 weeks to form tuber of potato on the node of the sterile shoot.</p> |
