NEW MATERIAL:Chitosanase having the following physico-chemical proprieties. Action; acting on chitosan and hydrolyzing beta-1,4-bonds. Substrate specificity; acting on chitosan or colloidal chitosan without acting on powdery chitin, colloidal chitin, glycol chitin, CMC and 1,4-polygalactosamine. Optimum pH; 6.0 (40 deg.C). Stable pH; 5-8 (40 deg.C for 30min). Range of action optimum temperature; acting up to 60// deg.C with 55 deg.C optimum temperature. Thermal stability and stabilization; stable at <=50 deg.C by heat treatment at pH6 for 15min, completely inactivated at 90 deg.C and increasing residual activity with increasing salt concentration within the range of 0.1-2.0 mol NaCl concentration in heat-treating at pH6 and 50 deg.C for 15 min. Molecular weight; 31000, etc. USE:An enzyme. PREPARATION:A strain of a microorganism No. K-881 belonging to the genus Bacillus (FERM P-10257) is cultured at 27-32 deg.C and initial pH 6.0-7.0 under aerobic conditions for 2-5 days. |
