摘要 |
PURPOSE:To produce a yeast catalyzing a metabolic reaction of mephenytoin useful as an antiepileptic agent using a genetic engineering technique by integrating a human-originated human liver cytochrome P-450MP into a plasmid. CONSTITUTION:A clone (lambda hPA6) containing cytochrome P-450 gene is separated from a human liver cDNA library to obtain human liver cytochrome P-450MP. A coding region except for a part of 5' terminal is sectioned from a plasmid containing a cDNA coding the human liver cytochrome P-450MP and the obtained coding region is linked to a separately chemically synthesized synthetic DNA sto reconstitute the whole base sequence. The obtained gene is integrated into a yeast expression vector to obtain a plasmid for expression. |
