PRODUCTION OF PHOSPHOLIPASE AZ OF HUMAN PANCREAS

PURPOSE:To make possible of mass production of human pancreas phospholipase Az, essential enzyme for structural analysis of phospholipid important as principal component of biomembrane and useful for diagnosis of pancreas disease by using gene recombination technique. CONSTITUTION:Following processes are performed to afford the aimed article: (a) nRNA is separated from human pancreas and cDNA is synthesized from said RNA, then cDNA library is prepared; (b) cDNA clone having gene coding human pancreas phospholipase Az is screened from said cDNA library; (c) a gene coding human pancreas phospholipase Az contained in said cDNA clone is inserted into plasmid vector and recombinant plasmid is constructed; (d) said recombinant plasmid is introduced into host cell and the host cell is transformed; (e) resultant transformant is cultured and (f) the aimed article is separated from the culture solution and collected.
