PURPOSE:To dye protein in high sensitivity in a short time, by immersing a substrate containing protein in a hemin solution so that the protein is changed into protein having peroxidase action, immersing the protein in a specific solution so that a dyestuff is synthesized on the separated protein. CONSTITUTION:Blood protein separated by two-demensional electrophoresis using polyacrylamide gel as a substrate is immersed in a buffer aqueous solution containing hemin, hemin is adsorbed on the protein, and the protein is converted into protein having peroxidase action. The substrate is immersed in a buffer solution containing no heim, hemin in the parts of the substrate having no protein is removed, the substrate is immersed in a buffer aqueous solution containing a phenol derivative (e.g., 2,6-dimethylphenol), a benzidine derivative (e.g., 3,3'- diaminobenzidine) and H2O2, a dyestuff is synthesized on the molecules of separated protein and colored. |
